filler for hollow breasts

If I was to fill the hollow portion of the large breasts of a doll with something like distilled water would this add to the feel or possibly ruin them?

It would certainly lead to long term issues. TPE is porous remember and should not really be in long term contact with any liquid that might soak into the material.

The biggest issue is that you would be putting over 20 lbs of water into a rubber balloon... that isn't as strong as a latex balloon.
So, if you stand her up, her breasts may rip off.
TPE is designed to be soft and squishy... which makes it the opposite of strong and supportive.
(you choose one aspect of a plastic, which usually comes at the cost of other aspects)

We are truly in an age of doll expansion. Sure it's the wild west, with each manufacturer trying to out do one another and society still poo-pooing dolls, but eventually, the age will mature, order will be found and one day we can bring out ladies anywhere.
